In vitro study demonstrates sustained 12-hour amlodipine release using high-viscosity polymers in effervescent floating tablets, suggesting improved gastric residence and drug bioavailability.
ABSTRACTGastric-retentive amlodipine besylate effervescent tablets have been developed using avariety of hydrophilic polymers. Formulations were developed using different grades ofdifferent concentrations of polymer. H. HPMC K4M, HPMC K15M, HPMC K100 andxanthan gum were used as polymeric substances. The formulated mixtures have beensubjected to various pre-formulation studies and flow properties, and all formulations showthat the powder mixtures have good flow properties. Among all formulations, drug releasewas delayed for the desired period, H. 12 hours, for the HPMC K100 formulation as apolymer. The dissolution data showed that formulations made with HPMC K100M andxanthan gum as the polymer sustained drug release for the desired period. H. 12 hours at aconcentration of 150 mg. On the other hand, in formulations containing HPMC K4M, HPMCK15M as polymer failed to produce the desired drug release. Therefore, the floating drugdelivery system of amlodipine using the appropriate amount of appropriate polymer canincrease the activity of the drug by prolonging the gastric residence time or decreasing thefloating lag time.
