The design features of an experimental transmission electron microscope employing an ultra high vacuum column, field emission gun and superconducting objective lens are described. The limitations on observations of the information in images at a level required for the determination of molecular structure of biogenic macromolecules are set by the characteristics of the biological specimen and the “noise” inherent in specimen preparation as well as the instrumental imaging and recording capabilities. These several factors are discussed.
