Review reveals broad antimicrobial and anticancer potential of novel 1,3-thiazinan-4-one hybrids, indicating their value as versatile scaffolds for drug discovery.
The 1,3‐thiazinan‐4‐one scaffold is a privileged six‐membered sulphur‐ and nitrogen‐containing heterocyclic framework with distinctive electronic properties, conformational adaptability, and hydrogen‐bonding capabilities, making it an attractive pharmacophore in medicinal chemistry. Hybrid molecules incorporating the 1,3‐thiazinan‐4‐one scaffold have emerged as promising medicinal chemistry candidates because of their structural diversity and broad biological activity. This review summarizes recent literature (2020–2026) in the synthesis of 1,3‐thiazinan‐4‐one hybrids bearing diverse heterocyclic motifs and their therapeutic potential. This scaffold has demonstrated therapeutic potential, exhibiting antibacterial, anticancer, antifungal, and antimicrobial activity, although many remain at the in vitro or preliminary biological evaluation stage. Structure–activity relationship (SAR) investigations have further revealed important structural determinants for enhanced biological efficacy. The review provides a focused and updated analysis of 1,3‐thiazinan‐4‐one hybrids incorporating diverse heterocyclic scaffolds, with emphasis on recent synthetic strategies, structural diversification, therapeutic activities, and emerging structure–activity relationships.
