This study describes nucleolytic activity of a copper‐metalated, multisite‐coordinating, hybrid polymer CPPL‐Cu under oxidative conditions. Rapid relaxation of supercoiled plasmid DNA was observed and mechanistic probing by chemical and enzymatic assays revealed involvement of singlet‐oxygen‐derived reactive species.
