After oral administration to human subjects, benorylate, 4-acetamido-phenyl-2-acetoxybenzoate, is well absorbed (83%) from the gastro-intestinal tract.2. Metabolism of benorylate yields salicylic acid and paracetamol in the blood and subsequent elimination in the urine follows previously described routes for these latter two compounds.3. The slower rate of absorption of benorylate compared to aspirin and paracetamol permits a less frequent dosage schedule to be used during multiple medication and additionally provides higher blood levels of active metabolites overnight.4. Two alternative formulations of benorylate, a suspension and a ‘Caplet’ were shown to have equivalent biological availabilities.
