Recent data demonstrate that sera from patients with Guillain–Barré syndrome react with ganglioside complexes (GSCs) consisting of two different gangliosides, but not with each constituent ganglioside. GSCs are likely to express conformationally different glycoepitopes from those of constituent gangliosides. Anti-GSC antibodies are real examples indicating that clustered glycoconjugates in the cell membrane are involved in antibody-mediated events. As well as increasing the spectrum of antiganglioside antibodies in Guillain–Barré syndrome and enhancing their value as diagnostic markers, the discovery of anti-GSC antibodies provides new vistas in antibody–antigen interaction through clustered carbohydrates on the cell membrane. This review illustrates a new paradigm for antibodymediated events through GSCs.
