An immunochemical assay method for bovine parathyroid hormone utilizing quantitative complement fixation is described in detail. Compared with biological assay methods generally and, specifically, the Munson method (serum calcium level in acutely parathyroidectomized rats), the new immunoassay method is more precise (λ =0.11), it is simpler to perform, and it can measure lower concentrations of hormone—as little as 24 rang (84 mU) /ml. Immunoassays and bio-assays of parathyroid hormone preparations of widely varying potency were in good agreement. Following storage at – 20 C in dilute neutral solution (<1 μg/ml), a decrease in antigenic activity of parathyroid hormone, thought to represent aggregation, was found to be reversible by brief sonic oscillation.
