A staining method for demonstrating Pneumocystis Carinii and fungi in unfixed specimens is described. The method is similar to Grocott's modification of Gomori's methenamine silver (GMS)method,but periodic acid is substitutedin place of chromic acid and very dilute ammoniacal silver in place of methenamine silver. Dilute ammoniacal silver is much more stableat high temperatures than methenamine silver and, therefore. is suitable for use in a microwave oven.The staining results are consistent and reliable and are indistinguishable from those obtained with Grocott's method. P. cariniiare easily identified because of nuclear-like staining, which may be sporozoites, often seen within the cysts. (The J Histotechnol 11: 19, 1988.)
