The local conformation of confined DNA is studied by emission polarization anisotropy. The DNA backbone aligns more in a narrower nanochannel displaying a larger difference in emission intensity polarized parallel and perpendicular to the extension axis (see image). This method is expected to provide useful information on local conformational changes of DNA in protein–DNA interactions, DNA compaction, and fundamental polymer physics in confined environments.
