The src gene of nondefective Rous sarcoma virus (ndRSV) encodes a protein that is responsible for transformation of cells. This protein has been identified by in vitro translation of 21S-24S virion RNA (Purchio et al. 1977; Brugge and Erikson 1977) and by immunoprecipitation of [35S]methionine-labeled polypeptides from RSV-infected chicken cells with an antisera raised in rabbits bearing RSV-induced tumors. The in vitro translation product and the polypeptide immunoprecipitated from RSV-infected cells are identical by two-dimensional peptide mapping. Both polypeptides have a molecular weight of 60,000. The src-gene product has been designated pp60src by Erikson and coworkers. This protein is not immunoprecipitated by antibodies directed against the viral structural proteins, polymerase, or envelope glycoprotein and is not immunoprecipitated by normal rabbit serum.
