Diagnosis in advance of symptoms i very useful for disease prevention and as a guide for instituting effec-tive therapeutic regimens. In principle, linkage meth-ods should become applicable to virtually any inher-ited disorder, given an adequate supply of genetic markers. Among such genetic markers are restriction-fragment-length polymorphisms (RFLPs) (Botstein et al. 1980), regions of variation of DNA sequence among individuals, which are commonly detected using radio-actively labeled cloned human DNA probes on gel transfers of human genomic DNA cleaved with restric-tion enzymes. These RFLP loci behave as codominant Mendelian alleles and thus can be used in family stud-ies to trace the inheritance of traits that cosegregate with a RFLP locus, even though almost nothing else
