Making circles: Nature solves the problem of peptide cyclization with the help of large modular synthetases that catalyze both the synthesis of the precursor peptide and its cyclization. Recently it was found that the isolated thioesterase domain of tyrocidine synthetase mediates the formation of cyclic peptides (such as 1) independently of the rest of the enzyme. This account compares the various biocatalytic approaches to peptide cyclization.
