Tissue homeostasis is dependent upon essential interactions between cells and the surrounding extracellular matrix. Within the skin, crosstalk between epidermal and dermal layers as well as matrix composition, can dictate the outcome of several dermatological pathologies, such as wound healing and psoriasis. Within this review we will highlight examples of keratinocyte–fibroblast crosstalk and cell interactions with the extracellular matrix, while emphasizing the importance of establishing coculture and 3D biomimetic models to better address pathophysiological processes in vitro.
