Dorsal ovariectomy before elastase infusion prevents adhesions and improves experimental AAA induction in female rats.
Why the study?
Women have increased abdominal aortic aneurysm growth rates and higher rupture risk, but the underlying hormonal effects remain unclear.
This protocol provides a reproducible method for dorsal ovariectomy and elastase-induced AAA in female rats, avoiding adhesions associated with ventral approaches to better study hormonal effects on AAA.
Women have increased abdominal aortic aneurysm (AAA) growth rates and a higher rupture risk, yet hormonal effects remain unclear. We present a protocol combining dorsal ovariectomy with elastase-induced AAA formation in female rats. We describe bilateral dorsal ovariectomy, a 42-day hormone depletion period, and ventral elastase infusion for AAA induction. This approach prevents adhesions encountered with traditional ventral techniques, increasing experimental feasibility and success. Zaghloul et al. (2025) studied Abdominal aortic aneurysm (AAA). Dorsal ovariectomy and elastase-induced AAA formation vs. Sham surgery was evaluated on Successful AAA induction (increase in aortic diameter). This protocol describes a dorsal ovariectomy approach combined with elastase-induced AAA formation in female rats to prevent adhesions and increase experimental feasibility.
