Reduction-controlled hierarchical unpacking is proposed by Z. Gu and co-workers on page 1534 for the development of virus-mimicking gene carriers. By combining a diselenide-conjugated oligoethylenimine/DNA inner core with a disulfide-modified hyaluronic acid shield, the resulting ternary polyplexes are designed to be stepwise triggered by the gradient glutathione (GSH) level both at the tumor site and inside the cells. The encouraging performance highlights the approach for more-efficient gene delivery.
