One tag, two labels: Defects in protein prenylation lead to a number of serious diseases; here we show that an azide-tagged substrate enables multilabel detection of defective protein geranylgeranylation by two geranylgeranyl transferases. Rapid in-gel visualization and affinity purification have been demonstrated on recombinant proteins, cell lines, and mammalian disease models.
