Two isolates of Entodinium longinucleatum have been cultured in vitro for at least six months in the presence of bacteria on a reduced buffered salts medium with the daily addition of starch grains and ground dried grass. Cultures of Entodinium longinucleatum were normally maintained by dilution, twice each week, with an equal volume of fresh medium, but the protozoa would also grow from a 2.5% inoculum with a mean generation time of 1.3 days.
