SummaryAn S25-RNase gene in the pistil responsible for self-incompatibility within the apple (Malus × domestica Borkh.) was cloned from a McIntosh cultivar. We developed an S25-allele-specific polymerase chain reaction and digestion (PCR-digestion) method using specific primers and the restriction enzyme BamHI. We investigated the S-allele genotypes of 18 ‘McIntosh’ progeny.
