The replication kinetics of liver cells have been studied in 1-day-old and in 3-week-old rats, at 3 and 5 weeks, respectively, after 2 and 10 μCi of H³ TDR per gram of body weight. Estimates of the respective generation times and their component intervals have been made from the changing percentages of labeled mitoses. In the younger rats, after 2 μCi/gm, the rhythm of replication has been disorganized. In the older rats a new rhythm has been established. The generation time of the replicating diploid hepatocyte has been doubled without change in either G2 + mitosis or DNA synthesis time intervals. There is marked prolongation of the G1 interval. At these levels of H³ TDR administration no histological abnormalities are found in the liver.
