The rate of dissolution of a relatively insoluble material deposited in the lower respiratory tract (LRT) is an important parameter in determining its relative retention in the LRT and its translocation to other tissues. Dissolution conditions may affect the rate of dissolution of a relatively insoluble material. The effects of some of these factors on the in vitro rates of dissolution of some radioactive aerosol particles were studied. Included among these factors were particle size, solvent composition, solvent flow rate, temperature of the dissolution system and method of measurement. Experimental conditions for the measurement of the in vitro rates of dissolution of relatively insoluble aerosol particles for comparison with in vivo dissolution rates in the LRT are discussed.
