Acid phosphatase was localized in Asteromonas gracilis (Chlorophyceae, Volvocales) using lead-salt and azo-dye cytochemical techniques. β-glycerophosphatase was partially characterized biochemically to determine an optimum method for Gomori-type (lead-salt) localization of that enzyme. A modified procedure for β-glycerophosphatase localization, based on the biochemical data and the results of other cytochemical methods, was employed. β-glycerophosphatase reaction product appeared in dictyosome-associated vesicles, provacuoles, and vacuoles. Naphthol AS-BI phosphatase (azo-dye) was localized in membranous coils within vacuoles. These results are compared with similar studies in other algae and vascular plants. Hydrolase compartmentation and the process of autophagy in this wall-less unicell are discussed.
