The binding of noncognate tRNAs to isoleucyl-tRNA synthetase is enhanced in mixed solvents. If the noncognate tRNA is acylated, isoleucyl-tRNA synthetase may hydrolyze the aminoacyl-tRNA bond. This reaction has several uses in the study of synthetase specificity.
