Isolated rat pancreases were perfused in an organ perfusion system with a medium containing 4.6 % dextran as the colloid and approximating plasma in its electrolyte composition. A glucose pulse of 2-min duration (producing a glucose concentration of 200–220 mg/100 ml) consistently stimulates insulin secretion if the perfusing medium contains Ca at 2.5 mEq/1. The secretory response is attenuatedif Mg is added to produce a concentration of 2.5 mEq/1 and is abolished when the Mg concentration reaches 5 mEq/1. With a Mg concentration of 5 mEq/1 secretion occurs if Ca also is elevated to 5 mEq/1. Further elevation of the Mg concentration to 15 mEq/1 again abolishes secretion with this higher Ca concentration. (Endocrinology85: 594, 1969)
