Osteoarthritis (OA) comprises joint and bone ailments aiding numerous pathological and molecular events constituting articular cartilage degeneration grievance and synovial and subchondral bone remodelling. Combating OA by various phenomena including pain killer therapy, surgeries, and drug delivery to the intra-articular joint space establishes temporary relief due to numerous microenvironmental changes such as dense tissue and negatively charged extracellular matrix. Recently, a substantial increase in both the conventional and contemporary levels of OA has been observed. Current clinical practices are chiefly confined to symptomatic management but are unable to bother the inability to prevent OA. In this silhouette, we discuss the nanogels and associated drug delivery-based carrier system for diagnosing, managing, and treating OA from the molecular level to the pathological level, owing to the exploration of integrated therapeutic molecules and regenerative medicine. Nanogel (NGs)-based drug delivery systems have established targeted molecular pathways and pathogenic mechanisms associated with OA progression. Owing to the targeted drug delivery, we explored NGs for the promotion of cartilage repair, intra-articular drug delivery, cell, gene, and biological molecule delivery of cargo for the next-generation OA therapeutics.
