A novel high-performance liquid chromatographic method has been developed for the determination of stevioside in the leaves of the plant Stevia rebaudiana (Bertoni). An isocratic water–acetonitrile eluent was employed with a LiChrosorb NH2 bonded silica column. The effluent was monitored by integrated pulsed amperometry in an alkaline medium (pH > 12) after adding sodium hydroxide post-column. Peak responses were linear from 1 to 100 mg L–1, with a limit of detection of 0.3 mg L–1. This method gave greater sensitivity and selectivity than UV detection.
