Unlike traditional cytotoxic drugs, CDK4–CDK6 inhibitors arrest progression through the G1 phase, promoting transient cell-cycle withdrawal into a quiescent state or, possibly, permanent proliferative arrest (senescence), either of which could avert tumor progression.
