Isolated pancreatic islets from normal, hypophysectomized, and hypophysectomizedrats treated with human placental lactogen (HPL) were studied in vitro. The release, content and synthesis of insulin were measured after incubation in media with either alow (60 mg/100 ml) or a high (300 mg/100 ml) glucose concentration. The 3 parameters were decreased in islets from hypophysectomized animals as compared with the normals. Theislets of hypophysectomized rats treated with HPL for 4 days (2 mg/animal/day) responded to glucose stimulation by an increased release and synthesis of insulin of the same magnitude as the normal controls. These results indicate that HPL facilitates the response of the islets to glucose stimulation and that this effect is independent of growth hormone. (Endocrinology84: 619, 1969)
