Multiple myeloma (MM) has the highest incidence of bone involvement among malignant diseases. 1 Multiple myeloma bone disease (MMBD) is characterized by osteoclast (OCL) activation with suppressed or absent osteoblast (OBL) function resulting in local bone destruction without new bone formation. 2 IL-3 is a bifunctional cytokine that is upregulated in MM patients, increases myeloma cell and OCL proliferation and indirectly suppresses osteoblastogenesis via CD14 + BM monocytes (BMM). 3 , 4 However, the monocyte-/macrophage-derived mediators of IL-3’s effects in MMBD are unknown.
