BACKGROUND: Autoimmune reactions, environmental factors, and heredity all have a role in the development of rheumatoid arthritis (RA). This article describes the role of gasdermin d (GSDMD) gene polymorphism in the pathogenesis of this disease.METHODS: Ninety volunteers were included in this trial; 20 patients had just been diagnosed with RA, 40 received treatment with methotrexate and Enbrel (etanercept), and 30 were healthy controls (20-60 years old). Genomic DNA was extracted from the blood sample using the EasyPure® Genomic DNA Kit. The GSDMD SNP (rs375764221) was detected using high-resolution melting (HRM) real-time PCR.RESULTS: There was notable elevation in the frequency of the AA genotype and the mutant allele (A) in newly diagnosed patients as compared to treated and control groups. Still, there was no significant difference in the mutant genotype (AA) distribution between the control and treated groups. There was a highly significant difference in the distribution of these genotypes among disease activity levels in the newly diagnosed RA group, there were no significant differences in the distribution of GSDMD genotypes among three disease activity levels in the treated RA group.CONCLUSIONS: The investigation’s findings demonstrate the part that genetic factors play in the development and progression of RA. The study of the GSDMD gene polymorphism is an area full of promise for significant discoveries on the mechanisms behind autoimmune diseases.
