HSPB8 p.Gly62Asp is associated with myopathy via post-translational modifications, while p.Lys141Glu disrupts dimerization and affects stress granule dynamics.
HSPB8 p.Gly62Asp is identified as the first N-terminal variant associated with myopathy, with p.Lys141Glu likely affecting stress granule dynamics.
Hypothesis-generating for HSPB8 N-terminal variants in myopathy; requires validation before diagnostic or research adoption.
Objectives: disorder. Methods: We reviewed clinical and laboratory data and performed molecular dynamics simulations to explore variant effect. Results: pathogenic variant, c.421 A>G (p.Lys141Glu). Molecular dynamics simulations suggested that p.Gly62Asp may exert its effects through post-translation modifications while p.Lys141Glu may disrupt dimerization. Discussion: HSPB8 p.Gly62Asp is the first N-terminal variant associated with myopathy. TIA1 aggregates, more prominent than Z-disk myofibril aggregates, suggest that p.Lys141Glu may affect stress granule dynamics more than Z-disk integrity.

Putko et al. (2025) studied HSPB8 Myopathy and Distal Motor Neuropathy. HSPB8 variants (p.Gly62Asp and p.Lys141Glu) was evaluated on Variant effect on protein function and pathology. HSPB8 p.Gly62Asp is associated with myopathy via post-translational modifications, while p.Lys141Glu disrupts dimerization and affects stress granule dynamics.
