Carbonic anhydrase activity of insect (cricket) testes homogenates can be demonstrated, after electrophoresis, in polyacrylamide gels buffered at pH 8.1 with Veronal to which a few drops of 0.2% bromthymol blue in 50% ethanol have been added. A distinct yellow band appears within 15-30 sec in the otherwise blue-tinted gel when it is placed in cold CO2 saturated water. Controls consisting of gels containing the separated enzyme gave negative results when placed in cold CO2-free water, CO3-free water buffered at pH 4.5, and CO2-free water at pH 2.9.
