This graphical review examines the evolution of paullones, a class of nitrogen-based chemical structures, from their origins as cancer-research tools to their current role as potential treatments for Alzheimer’s disease. These compounds are characterized by a unique seven-membered ring architecture that allows them to effectively block enzymes. Herein, various chemical synthesis methods are presented, highlighting a shift from traditional techniques to modern, environmentally friendly protocols that do not require heavy metals. By analyzing structure-activity relationships, this graphical review shows how specific molecular modifications can enhance a drug's ability to target multiple biological pathways simultaneously.
