Aiming at the 170-year-old puzzle of the origin of molecular chirality in chemistry, this paper constructs subjectivity-free mathematical equations for theoretical derivation based on the ontological principle of matter, the natural canonical ratio (equilibrium ratio), and the proportional constraint rule of material primordium. It is verified that molecular chirality and enantiomeric differentiation do not arise from external perturbations such as symmetry breaking, weak interactions or circularly polarized light, but are inevitable outcomes governed by the intrinsic proportional mechanism of matter. Existing theories can describe chiral amplification processes yet fail to explain the origin of chiral configurations. The equations in this paper introduce no extra fitting parameters, and derived results are in good agreement with measured data from stereochemistry, crystallography and biomolecular experiments. This paper systematically analyzes the inherent defects of three mainstream theories, clarifies the intrinsic nature of chiral phenomena, and elaborates the mechanism for the stable existence of two enantiomeric configurations and the formation of homochiral preference in systems. It fills the core theoretical gap in stereochemistry and the study of the origin of life, and provides new theoretical support for relevant research directions.
