While intercepting Type 1 Diabetes prior to clinical onset remains challenging, early intervention represents a critical therapeutic frontier. In T1D, the immune system fails to maintain central and peripheral tolerance, targeting and selectively destroying pancreatic beta cells. Nearly 9.5 million people worldwide live with T1D, and India's burden is growing especially fast, at 3-5% each year. Aim: This study evaluates how biomaterial-based tolerogenic nanoparticles engineered to deliver pancreatic autoantigens alongside immunomodulatory cues can drive targeted expansion of functional FOXP3+ Tregs. This case study of distinct nanoparticle platforms indicates that pairing Treg biology with precision nanotechnology demonstrates the potential of "Inverse vaccines" as a viable, non-immunosuppressive disease-modifying therapy for T1D.
