GFPuv is a UV-excitable fluorescent reporter that enables rapid monitoring of gene expression, particularly in microbial systems. However, its application in filamentous fungi remains limited. Here, we evaluated GFPuv variants and identified one that performs robustly across Aspergillus species and the yeast Yarrowia lipolytica. Under UV illumination, transformed green fluorescent colonies can be visualized without colony dissection or microscopy, significantly simplifying screening. CRISPR-based genome editing has not been previously established in Aspergillus pseudoterreus. By integrating the optimized GFPuv reporter into a transformation and selection system, we established, to our knowledge, the first CRISPR knockout platform in this species. Targeted disruption of the genes pyrG, CAD, and PrID505606 validated the platform. PCR and Sanger sequencing confirmed that GFPuv-positive colonies were true mutants, while loss-of-function phenotypes of pyrG and CAD further supported platform reliability. This work establishes GFPuv as a rapid, microscopy-independent screening tool in select fungi and provides a foundational CRISPR framework for engineering A. pseudoterreus.
