This research includes the synthesis of chalcones due to their importance in organic chemistry; certain intermediates are used to form heterocyclic. Firstly, synthesis compound A1 was synthesized by the reaction between Benzo[d][1,3]dioxol-5-amine and chloroacetyl chloride. Then, cyclization by ammonium isothiocyanate is performed using acetone as a solvent to get compound A2. This compound contains an activated α-H that reacts with sub. Aldehyde, also glacial acetic acid & sodium acetate to yield (A3-A6). It can cyclize to fused heterocyclic compounds by reacting with hydrazine hydrate to obtain (A7-A10). Finally, to form (A11-A14) pyrimidin-thione derivatives, compound (A3-A6) was reacted with thiourea. Identification of (A1-A14) by spectroscopy (IR, H-NMR) was tested; their bioactivity against two types of bacteria, E. coli and S. aureus. Compounds A10,A11,A12 exhibited considerable inhibitory activity against S. aureus, with inhibition zones of (44, 32, 36) mm, respectively. In contrast, the antibacterial assay against E. coli showed that compounds A11, A12 produced inhibition zones of 22 and 20 mm, respectively. These findings indicate that the tested compounds exhibited variable antibacterial activity depending on the bacterial strain.
