The biochemical categorization of N,N-Dimethyltryptamine (DMT) strictly as an exogenous psychotomimetic has been superseded by empirical evidence demonstrating its role as an intrinsic mammalian neuromodulator and cellular chaperone. This paper establishes the comprehensive neurobiology, receptor kinetics, and biophysical mechanisms underlying the human "Biological Interface". First, we review the two-step enzymatic cascade governing endogenous biosynthesis, wherein aromatic L-amino acid decarboxylase (AADC, with cofactor pyridoxal 5'-phosphate) and the rate-limiting enzyme indolethylamine N-methyltransferase (INMT, utilizing S-adenosyl-L-methionine) convert dietary L-tryptophan into DMT. We detail tissue-specific localization beyond the pineal gland—including robust transcriptional expression across the cerebral cortex, choroid plexus, and peripheral lungs—and examine in vivo microdialysis data demonstrating acute, multi-fold surges of cerebral DMT during lethal hypoxia and cardiac arrest that match or exceed baseline monoamine concentrations. Second, we evaluate the differential receptor kinetics of DMT across transmembrane G-protein coupled receptors (\(5-HT{2A}\), \(5-HT{1A}\), \(5-HT{2C}\)) and the intracellular Sigma-1 (1) chaperone receptor residing at the endoplasmic reticulum-mitochondria-associated membrane (ER-MAM). We characterize the molecular mechanics of functional selectivity (biased agonism), contrasting canonical \(G{q/11}\)-phospholipase C signaling against non-canonical -arrestin-2 recruitment and immediate early gene transcription (Egr-1, Egr-2, c-Fos). At the 1 locus, DMT binding drives dissociation from BiP/GRP78 to stabilize inositol 1,4,5-trisphosphate receptor type 3 (IP3R3), preserving regulated mitochondrial Ca2+ influx, suppressing reactive oxygen species (ROS), and activating anti-apoptotic cascades (Bcl-2). Concurrently, psychoplastogenic signaling through TrkB autophosphorylation and BDNF upregulation drives rapid spinogenesis, dendritic arborisation, and enhanced synaptic efficacy. Third, we analyze the multi-compartmental pharmacokinetics of Target-Controlled Intravenous Infusion (TCI), demonstrating how continuous steady-state titration overrides rapid monoamine oxidase A (MAO-A) clearance to sustain an unconstrained cognitive plateau suitable for functional neuroimaging. Under sustained saturation, BOLD fMRI and high-density EEG exhibit profound Default Mode Network (DMN) disintegration, the collapse of cortical alpha (8–12 Hz) oscillations, and an exponential increase in global functional connectivity and Lempel-Ziv network entropy. Fourth, we synthesize these findings into the Transceiver Architecture, incorporating Jacobo Grinberg’s Syntergic theory, the piezoelectric resonance of pineal calcite microcrystals, and ventricular acoustic standing waves, illustrating how the breakdown of hierarchical predictive coding (REBUS model) enables the biological chassis to function as a direct field transducer. Finally, we trace the deep phylogenetic conservation of 5-HT2A, 1, and astrocytic cannabinoid receptor networks, resolving the evolutionary "lock-and-key" paradox and demonstrating that legislative prohibition constitutes a structural suppression of endogenous cellular neuroplasticity and sovereign human potential.
