Protein p66Shc is the heaviest isoform of the SHC1 gene. According to modern data, p66Shc is involved in many complex physiological and pathological processes in the body – in aging, oxidative stress, endothelial damage, atherogenesis. However, understanding its role in carcinogenesis is of particular interest. The involvement of p66Shc in the development of pancreatic cancer, esophageal cancer, thyroid cancer, ovarian cancer, colorectal cancer, and lung cancer has been demonstrated. It has been established that p66Shc can act as a suppressor and a stimulator of carcinogenesis in various types of tumors. The p66Shc protein is involved in the regulation of apoptosis, anoikis, and the modulation of reactive oxygen species production and autophagy, and its role in the pathogenesis of malignant tumors continues to be actively studied. In this regard, it is relevant to analyze the literature data on the role of the p66Shc protein in the pathogenesis of malignant tumors. To prepare the review, the Scopus, Web of Science, Medline, PubMed, CyberLeninka, RSCI, and CNKI databases were searched. Sources indexed in the Scopus, Web of Science, and PubMed databases were used for the analysis, and 14% of the studies were published in the last 5 years. 49 sources were used to write this literature review.
