The ethanolic extracts obtained from the aerial parts (EPE-aerial) and roots (EPE-root) of Jordanian Echinops polyceras were assayed for their TPC, TFC, antioxidant capacity using different assay methods, in addition to targeted LC-MS/MS analysis. EPE-root extract had higher TPC (92.31 mg GAE g−1), DPPH•, ABTS•+ (43.81; 108.19 μg mL−1, respectively) and FRAP (173.21 mg TE g−1) activities. EPE-aerial extract had higher TFC and CUPRAC activity (672.50 mg QE g−1; 227.35 mg TE g−1, respectively). Targeted LC-MS/MS analysis revealed each of CAPE (1151.6 mg kg−1) and hesperetin (1015.0 mg kg−1) as the main constituents in EPE-aerial and EPE-root extracts, respectively.
