This review aims to: (a) highlight key strategies for preparing non-racemic polythiophenes as single chemical entities or supramolecular thin-film architectures, (b) discuss their essential structural features and optical stability, and (c) overview their applications as chiral sensors, chiral catalysts, and optical materials. Special emphasis is placed on recent advances in the synthesis of chiral polythiophene, higher-order molecular organization driven by point, through-space, planar or axial stereogenicity, structure–property relationships, and the intended applications of these materials.
