Bacterial infections remain a significant public health challenge, with Staphylococcus aureus being one of the major pathogens responsible for skin, soft tissue, and respiratory tract infections. Long-term use of synthetic antibiotics may lead to adverse effects and accelerate the development of antimicrobial resistance, highlighting the need to explore alternative antibacterial agents derived from natural sources, such as Golkar (Chromolaena odorata L.). This study aimed to identify the phytochemical profile and evaluate the antibacterial activity of ethanol extract of Golkar leaves against Staphylococcus aureus. This research employed a true experimental design with a posttest-only control group design arranged in a Completely Randomized Design. The treatments consisted of ethanol extract of Golkar leaves at concentrations of 50%, 75%, and 100%, with chloramphenicol as a positive control and distilled water as a negative control. Phytochemical screening was conducted qualitatively, while compound identification was performed quantitatively using GC-MS analysis. Antibacterial activity was assessed using the diffusion method by measuring the inhibition zones of bacterial growth. The phytochemical test results indicated the presence of alkaloids, saponins, steroids, and terpenoids. GC-MS analysis identified several bioactive compounds, including eugenol, terpenoids, methyl palmitate, long-chain alcohols, aromatic esters, digitoxin, and gibberellic acid. The antibacterial assay demonstrated a concentration-dependent inhibitory effect, with optimal activity observed at concentrations of 75% and 100%. In conclusion, the ethanol extract of Golkar leaves shows potential as a natural antibacterial agent against Staphylococcus aureus.
