This project investigates rhizosphere bacterial communities associated with nitrogen-use-related alleles and their combinations in rice (Oryza sativa). The plant materials comprise the recurrent parent HJX74, four single-segment substitution lines carrying IAPAR9-derived alleles associated with OsWRKY23, DNR1, RNR10 and OsWRI1a, and five double-segment pyramiding lines. Plants were grown under no nitrogen addition (LN) and nitrogen addition (HN) conditions. A total of 60 rhizosphere soil samples were collected. Three separately collected pooled rhizosphere soil samples were obtained per genotype under each nitrogen condition, each comprising rhizosphere soil from five seedlings. The V3-V4 region of the bacterial 16S rRNA gene was amplified using primers 338F/806R and sequenced on the Illumina MiSeq platform with paired-end 300-bp reads. These data support comparisons of rhizosphere bacterial community diversity and composition among rice materials and nitrogen conditions, together with the identification of differentially abundant ASVs and candidates with predicted nitrogen-cycle-related functions.
