The advent of proline-based organocatalysis, pioneered by List and MacMillan, has established a sustainable, metal-free paradigm for asymmetric synthesis, complemented by advancements in peptide-based and supramolecular catalysts that enhance enantioselectivity. Concurrently, modern catalytic system, such as chiral copper and nickel complexes, have unlocked novel reactivity profiles, while heterogeneous catalysts have facilitated scalable and recyclable aldol methodologies. Innovations in reaction variants, including vinylogous and nitroso aldol reactions, alongside stereodivergent transformations, have broadened the scope of the asymmetric aldol reaction to address complex synthetic challenges. This review provides a comprehensive analysis of recent progress, focusing on catalyst design, mechanistic understanding, and the synergy between experimental and computational methodologies. By integrating established approaches with cutting-edge innovations, we elucidate the current state of the asymmetric aldol reaction and propose future directions for catalyst development and sustainable synthetic strategies.
