## Abstract Targeting mutant KRASG12C with covalent Switch-II pocket inhibitors (e.g., Sotorasib/AMG 510, Adagrasib/MRTX849) represents a major milestone in precision oncology. However, despite rapid initial objective response rates in non-small cell lung cancer (NSCLC) and colorectal cancer (CRC), clinical durability is strictly truncated by rapid adaptive resistance: median progression-free survival (PFS) plateaus at approximately 6.5 months. Standard clinical intuition attempts to overcome resistance by escalating drug dosage to achieve >95% target occupancy or by adding high-dose downstream inhibitors (e.g., MEK inhibitors). This strategy invariably collapses due to dose-limiting host toxicity, diarrhea, hepatotoxicity, and failure to halt wild-type RAS/RTK rebound. In this work, we demonstrate how the **Epistemic Life Engine** diagnoses and resolves this clinical impasse across two coupled scales: an 8-node systems oncology dynamical hypergraph ( β₁ = 5 ) and an explicit semiempirical quantum chemical reaction model (MOPAC v23.2.2 PM7): 1. **Systemic Feedback Paradox and Frustration Divergence (Φ = 439.73 100):** In an 8-node nonlinear ordinary differential equation (ODE) network modeling RTK, SHP2, SOS1, KRAS-G12C, wild-type RAS (NRAS/HRAS), MAPK (p-ERK), PI3K/AKT, and MYC survival drive, saturating Sotorasib monotherapy ($95%$ target occupancy) depresses p-ERK nadir to $0.5935$, which immediately relieves downstream DUSP6/SPRY negative feedback. Relieved feedback hyperactivates upstream Receptor Tyrosine Kinases (RTK, $0.2572$) and channels active nucleotide exchange via SOS1 into wild-type RAS ($0.5645$), driving full p-ERK rebound ($0.6047$) and sustaining tumor survival drive ($0.5277$). Because the local target gradient vanishes under saturating occupancy while residual oncogenic flux persists, the **Cognitive Frustration Index diverges to Φ = 439.73 τ = 100**, quantitatively signaling a futile in-system single-target trap.2. **Brute-Force Doublet Toxicity Failure (M(D) = 1.6881 > 1.0):** Combining high-dose Sotorasib ($90%$) with a MEK inhibitor ($85%$) suppresses p-ERK ($0.1956$) but drives upstream RTK hyperactivation ($0.5512$) and PI3K/AKT bypass ($0.5495$), while the super-linear **Metabolic Toxicity Tax** blows past the clinical ceiling (M(D) = 1.6881 > 1.0), predicting severe patient morbidity.3. **The Sub-Threshold Tri-Point Confluence Pinch (β₁: 5 → 2 → 0):** Rather than attempting single-node saturation, the Epistemic Engine deploys an algebraic network pinch that distributes low-intensity, sub-threshold inhibition across three confluent upstream funnel nodes: **$35%$ G12C + $30%$ SHP2 + $30%$ SOS1** coupled with a pulsatile temporal schedule (48h ON / 72h OFF). This topological intervention simultaneously chokes the nucleotide-exchange funnel into both mutant G12C and wild-type RAS, maintaining host safety (M(D) = 0.7221 ≤ 1.0), collapsing the Cognitive Frustration Index to **Φ = 0.55**, and driving Day-30 p-ERK to **$0.1582$** and phenotypic tumor survival drive down to **$0.1922$** (a $>70%$ reduction vs baseline and a $63.6%$ reduction vs monotherapy steady-state).4. **Quantum Chemical Warhead Pre-Organization (8.75\,kcal/mol Barrier Relief):** Using MOPAC PM7 reaction scans, we model the covalent nucleophilic Michael addition between the Sotorasib acrylamide warhead and the Cys12 thiolate (S⁻). In the unassisted Switch-II pocket, the activation enthalpy barrier is Δ H^ = 46.34\,kcal/mol. Upstream SOS1/SHP2 GDP-locking conformationally pre-organizes the pocket, positioning the catalytic Lys16 ammonium group to form a strong hydrogen bond (d(N-O) ≈ 2.75\,) with the acrylamide carbonyl oxygen, stabilizing the enolate-like transition state. This collapses the nucleophilic addition barrier to Δ H^ = 37.59\,kcal/mol—a barrier relief of **∇ 8.75\,kcal/mol** yielding an Eyring rate acceleration of **1.48 × 10⁶×** at 310\,K (37^). This multi-scale benchmark proves that synergistic, sub-threshold topological intervention preserves host tolerability while achieving superior oncogenic pathway silencing through simultaneous network cycle collapse and catalytic active-site pre-organization.
