Scaling adherent viral vaccine processes in roller bottles often increases operational burden in proportion to production capacity, with vessel count, manual handling, and staffing becoming limiting before cell biology does. This article describes a process redesign that combined a new cell line with a consolidated static culture platform, resulting in an approximately 20-fold increase in relative viral titer, a 60% reduction in operators (from five to two), an 86% reduction in units handled (from 400–57), a reduction in processing time from 4.5–1.5 h, and an antibiotic-free process, with animal and serological studies confirming that the final product maintained its expected characteristics and immunological response.
