Abstract

This protocol is for extraction of high-quality total RNA from Aiptasia larvae to use in NGS library preparations and RT-qPCRs.The protocol is written for the Zymo Research Quick-DNA/RNA Microprep Plus Kit (D7005) but can also be adapted for the Zymo Research Quick-RNA Microprep Kit, the additional reagents needed are listed.This protocol works well with ≥200 larvae between 1 dpf and 9 dpf and should produce ~100-500 ng total RNA, for older/smaller larvae, higher numbers might be required.
