目的 探讨乌梅丸(WMP)对肥胖小鼠血管内皮功能的作用及对氧化低密度脂蛋白(ox-LDL)诱导的血管内皮损伤的影响及机制。 方法 采用高脂饮食诱导肥胖小鼠模型,共喂养24周,随后将模型建立成功的小鼠随机分为模型组、乌梅丸低、中、高剂量组、二甲双胍组。对照组和模型组给予生理盐水灌胃,其余组给予相应的药物灌胃,共干预7周。记录体质量,采用HE染色观察主动脉形态学变化,检测血清血脂含量,ELISA法检测各组小鼠主动脉炎症因子水平。体外培养HUVECs,应用CCK-8法、TUNEL法、成管实验、划痕实验、Transwell实验分别检测细胞增殖、凋亡、成管、迁移能力。 结果 1)与模型组比较,乌梅丸高剂量组体质量、血脂、丙二醛含量下降(P0.05),过氧化氢酶水平上升(P0.01),白介素1β、白介素6水平降低(P0.001),HE染色显示主动脉组织形态得到改善;2)与ox-LDL组比,10%WMP组细胞增殖、迁移能力提升(P0.05),凋亡能力显著下降(P0.001)。 结论 乌梅丸可改善肥胖小鼠的血管内皮功能、减轻ox⁃LDL诱导的血管内皮细胞损伤,其机制可能是降低炎症反应、抗氧化,促进内皮细胞增殖迁移、抑制细胞凋亡。
