The Front Cover highlights advances in CRBN-recruiting molecular glue degraders since 2023, illustrating the shift from canonical glues to non-canonical chemotypes that expand the neosubstrate space. A glutarimide anchor binds to CRBN, while ligand-specific features shape the CRBN–ligand–neosubstrate interface. AI-enabled modeling can further accelerate the discovery of atypical MGDs.
