Abstract

目的 探讨黑果枸杞+肉苁蓉通过调控磷脂酰肌醇3-激酶(PI3K)/丝氨酸-苏氨酸激酶(Akt)信号通路对老年性骨质疏松症(SOP)大鼠的影响。 方法 48只SD大鼠随机分为空白组、衰老组、VE组(0.018 g·kg-1)、PI3K激活剂组(740 Y-P组,10 mg·kg-1)、黑果枸杞+肉苁蓉组(1 440 mg·kg-1)、黑果枸杞+肉苁蓉+740 Y-P组。Micro-CT检测大鼠骨微结构变化;HE染色观察股骨病理形态、碱性磷酸酶钙-钴染色检测大鼠股骨中成骨细胞活性、抗酒石酸酸性磷酸酶(TRAP)染色检测大鼠股骨组织中破骨细胞活性;ELISA法检测血清肿瘤坏死因子-α(TNF-α)、骨碱性磷酸酶(BALP)、抗酒石酸酸性磷酸酶5b(TRACP-5b)含量;Western blot检测股骨组织PI3K/Akt通路蛋白表达。 结果 与空白组比较,衰老组大鼠股骨微结构明显破坏,BMD、BV/TV、Tb.N、Tb.Th显著下降(PPPPP 结论 黑果枸杞+肉苁蓉可通过调控PI3K/Akt通路改善SOP。
