Here, we present a protocol for inducing focal striatal stroke in mice using photothrombosis. We describe steps for photothrombotic striatal surgery, magnetic resonance imaging, open field behavioral assessment of stroke-induced motor deficits, and post-stroke monitoring. We then detail procedures for tissue collection and immunofluorescence. This protocol provides a standardized approach for consistently modeling striatal stroke in mice, enabling mechanistic studies and preclinical therapeutic testing. • Standardized procedure for inducing focal striatal photothrombosis in mice • Steps for using mouse MRI to monitor and quantify stroke volume progression • Protocol for assessing mouse locomotion and behavior before and after stroke • Instructions for perfusion, sectioning, and immunofluorescent staining Publisher’s note: Undertaking any experimental protocol requires adherence to local institutional guidelines for laboratory safety and ethics.
